Each 4.4 g vial is enough to prepare 1 gallon of pH 7.15 buffer solution for use in the 5-step staining process. The buffer is helpful with the development of the correct, clear nuclear color schemes.

Directions:
Prepare five containers for the following dipping instructions.

  1. Fixative Solution 100%.
    Dip slide for a few seconds (air dry completely).
  2. Wright Stain 70% Fixative Solution 30%.
    Dip slide for 2 minutes.
  3. Wright Stain 20% Buffer Solution 80%.
    Dip slide for 5 minutes.
  4. Buffer Solution 16.7% Distilled Water 83.3%.
    Dip slide for 40 seconds.
  5. Buffer Solution 100%.
    Dip slide for 30 seconds.

General Notes
  1. When product/procedure work as expected at the beginning and then later results are not as high quality, change ALL solutions, as they are probably contaminated. It usually occurs during continuous, heavy use and when jars are left open (exposed) to the air. Exposure to moist air causes precipiation.
  2. Make sure all slides are clean prior to making the blood smear, if not, stain will not absorb properly onto the smear.
  3. Whenever automated staining machines are used, it is very important not to allow any chemical to dry in the delivery tubes. The lines utilized in steps 1, 2, and 3 must be rinsed with fixative (Methanol) and the lines used in steps 4 and 5 must be rinsed with distilled water to insure proper delivery of all process chemicals.
